The Lexus RX 350 was launched in January 2009 in Japan as a CUV (crossover utility vehicle) under the Lexus brand. The RX series had been introduced to the North American market in March 1998 as a new genre of crossover vehicle, and enjoyed enormous success. Although the Lexus RX 350 was the third-generation model, it was the first to be marketed in Japan under the Lexus brand, the first- and second-generation models having been marketed in Japan in 1998 and 2003, respectively, under the Toyota Harrier name.

Following the release of the RX 350 with a V6 3.5-liter engine (2GR-FE) in January 2009, the RX 450h hybrid with a V6 3.5-liter engine (2GR-FXE) was introduced in April, followed by the RX 270 with a 2,700 cc, inline 4-cylinder engine (1AR-FE) in August 2010.

The RX 350 and the RX 450h offered a choice between all-wheel-drive and front-engine, front-wheel-drive (FF) layouts, while only an FF configuration was available with the RX 270. The body dimensions were the same for all three models at 4,770 mm in length, 1,885 mm in height, 1,690 mm in width, and a wheelbase of 2,740 mm.
