The second-generation Toyoace was launched in March 1959. Retaining the semi cab-over-engine design while moving the engine backward to beneath the seats and adopting a column shifter, the second-generation model increased the passenger capacity from two to three. The body dimensions were also slightly increased, and the two-piece windshield was replaced with a single-piece windshield. Adopting a 2-tone color scheme, the vehicle made its standard body color brighter, while using a darker tone of the same color for the lower part of the cabin.

A tilt-cab mechanism was introduced to improve serviceability. The engine was initially a 1-liter 33 PS unit (Type S), which was replaced with a newly developed 1-liter 45 PS unit (Type P) in October 1959. The second-generation Toyoace added a 1.2-liter, 1.25-ton model and a 1.5-liter, 1.5-ton model, becoming a long seller that remained in production until 1971.
